BLOT Bordetella IgM

BLOT kit for the detection of specific IgM antibodies to the recombinant antigens of Bordetella pertussis and separated antigens of Bordetella parapertussis in human serum and plasma.

Description:

• Used antigens: extract of Bordetella parapertussis antigens rich in specific protein antigens (FHA, pertactin), enriched in selected parts of recombinant Bordetella pertussis antigens (pertussis toxin, FHA).
• Protein antigens of Bordetella parapertussis are separated by gel electrophoresis, and then transferred to the nitrocellulose membrane, recombinant antigens of Bordetella pertussis are transferred to the nitrocellulose membrane (i.e. to the respective WB strips).
• If specific antibodies are present in the sample, they will bind to the respective antigens.
• The complex is labelled with Conjugate and detected through a colour reaction with substrate (BCIP/NBT).
• The kit contains 16 tests.
• Short incubation periods, total assay time: approximately 1.5 h.
• High sensitivity and specificity.
• All reagents are supplied in working strength.
• The kit may be used sequentially for smaller batches of samples, reagents are provided in sufficient quantities.
• Strips are equipped with the control and CUT-OFF lines to check validity, functionality and sensitivity of the kits and for visual facilitation of test evaluation.
• Positive and Negative controls included enabling validation of the tests.
• Colour identification in compilance with antibody classes.
• Blotting trays included.
• Identical (i.e. interchangeable) reagents (except for Conjugate) in the BLOT Bordetella kiks.
• Evaluation of results – clear criteria using Evaluation table.

Applications:

• Diagnostic of pertussis and parapertussis, confirmative test

Brief assay procedure:

1. Dilute serum/plasma samples (1:50).
2. Pipette diluted samples and controls into the channels of the tray and insert the WB strips.
3. Incubate for 30 min at room temperature using a shaker.
4. Wash 3 times for 5 min.
5. Add Conjugate.
6. Incubate for 30 min at room temperature using a shaker.
7. Wash 3 times for 5 min.
8. Add substrate (BCIP/NBT).
9. Incubate for 15 min at room temperature using a shaker.
10. Wash twice for 5 min in distilled water.
11. Dry the strips and evaluate results using the validation strip enclosed or using IMUNOBLOT software.
